<li><strong>One-shot immunization with Sputnik Light (the first component of Sputnik V vaccine) is effective against SARS-CoV-2 Delta variant: efficacy data on the use of the vaccine in civil circulation in Moscow</strong> -

Objectives Vaccination remains the most effective response to the COVID-19 pandemic. Most vaccines use two-dose regimens. In turn, single-dose vaccines also have high potential, since, on the one hand, they simplify the vaccination program, make it more accessible and convenient for more people around the world, and on the other hand, they are better suited for subsequent revaccination. However, there is not enough data on the effectiveness of single-dose vaccine variants against new genetic lines to assess their current potential. It is not clear how much a single dose of immunization protects against the globally dominant delta variant. In this work, we investigated the effectiveness of a single dose vaccine (Sputnik Light, the first component of Sputnik V vaccine) against the Delta variant in Moscow. Methods To assess the effectiveness of one dose of viral vector vaccine based on rAd26 against the delta variant in Moscow, we used data from the Moscow registries of vaccination against COVID-19 and the incidence of COVID-19. The availability of data on the number of seropositive residents of Moscow made it possible to consider the size of the immune layer formed because of a previous COVID-19 disease or vaccination. To calculate the effectiveness, the proportion of COVID-19 cases among those vaccinated with a single dose and the proportion of cases among those who were not vaccinated in July 2021. Results Our data indicate that throughout July 2021, the dominant variant of the coronavirus at the level of 99.5% in Moscow was the SARS-CoV-2 delta variant and its subsidiary lines. Considering the immune layer of 46% allowed us to calculate the effectiveness of a one-shot vaccine against the delta variant in Moscow during the first three months after vaccination at the level of 69.85% (95% confidence interval [CI], 64.08 to 74.70). In the 18-29-year-old group, the overall vaccine efficacy against the delta variant was 88.61%, in the 18-59 group - 75.28%. Sputnik Light demonstrates higher efficacy against Delta variant than many two-shot vaccines. Conclusion The results indicate a high efficacy of a single immunization first component of Sputnik V vaccine against delta variant among young and middle-aged people (86.2% and 75.28%, respectively), at least during the first 3 months after receiving the one-shot vaccine.

Purpose: Long COVID syndrome is now a real and pressing public health concern. We cannot reliably predict who will recover quickly or suffer with mild debilitating long COVID 19 symptoms or battle life threatening complications. In order to address some of these questions, we studied the presence of symptoms and various correlates in COVID 19 patients who were discharged from hospital, 3 months and up to 12 months after acute COVID 19 illness. Methods: This is an observational follow up study of RT PCR confirmed COVID 19 patients admitted at 3 hospitals in north India between April August 2020. Patients were interviewed telephonically using a questionnaire regarding the post COVID symptoms. The first tele calling was done in the month of September 2020, which corresponded to 4 to 16 weeks after disease onset. All those who reported presence of long COVID symptoms, were followed up with a second call, in the month of March 2021, corresponding to around 9 to 12 months after the onset of disease. Results: Of 990 patients who responded to the first call, 615 (62.2%) had mild illness, 227 (22.9%) had moderate and 148 (15.0%) had severe COVID 19 illness at the time of admission. Nearly 40% (399) of these 990 patients reported at least one symptom at that time. Of these 399 long COVID patients, 311 (almost 78%) responded to the second follow up. Nearly 8% reported ongoing symptomatic COVID, lasting 1 to 3 months and 32% patients having post COVID phase with symptoms lasting 3 to 12 months. Nearly 11% patients continued to have at least one symptom even at the time of the second interview (9 to 12 months after the disease onset). Overall, we observed Long COVID in almost 40% of our study group. Incidence of the symptoms in both the follow ups remained almost same across age groups, gender, severity of illness at admission and presence of comorbidity, with no significant association with any of them. Most common symptoms experienced in long COVID phase in our cohort were fatigue, myalgia, neuro psychiatric symptoms like depression, anxiety, brain fog and sleep disorder, and breathlessness. Fatigue was found to be significantly more often reported in the elderly population and in those patients who had a severe COVID 19 illness at the time of admission. Persistence of breathlessness was also reported significantly more often in those who had severe disease at the onset. The overall median duration of long COVID symptoms was 16.9 weeks with inter quartile range of 12.4 to 35.6 weeks. The duration of symptom resolution was not associated with age, gender or comorbidity but was significantly associated with severity of illness at the time of admission (P=0.006). Conclusions: Long COVID is now being recognized as a new disease entity, which includes a constellation of symptoms. Long COVID was in almost 40% of our study group with no correlation to age, gender, comorbidities or to the disease severity. The duration of symptom resolution was significantly associated with severity of illness at the time of admission (P = 0.006). In our study, all patients reported minor symptoms such as fatigue, myalgia, neuro psychiatric symptoms like depression, anxiety, brain fog and sleep disorder and persistence of breathlessness. Severe organ damage was not reported by our subjects. This might be the longest post COVID follow up study on a sample of nearly 1000 cases from India.

The rapid spread of SARS-CoV-2 continues to impact humanity on a global scale with rising total morbidity and mortality. Despite the development of several effective vaccines, new products are needed to supply ongoing demand and the needs of specific populations. We report herein a pre-specified interim analysis of the phase 2 portion of an ongoing Phase 2/3, randomized, placebo-controlled trial of a coronavirus virus-like particle (CoVLP) vaccine candidate produced in plants that displays the SARS-CoV-2 spike glycoprotein adjuvanted with AS03 (NCT04636697). A total of 753 subjects were recruited between 25 November 2020 and 24 March 2021 into three groups: Healthy Adults (18-64 years: N=306), Older Adults (≥65 years: N=282) and Adults with Comorbidities (≥18 years: N=165) and randomized 5:1 to receive two intramuscular doses of either vaccine CoVLP (3.75 μg/dose + AS03) or placebo 21 days apart. This report presents safety, tolerability and immunogenicity data collected up to 21 days after the second dose. The immune outcomes presented include neutralizing antibody (NAb) titres and cellular (γ and IL-4 ELISpot) responses. In this study, CoVLP+AS03 was well-tolerated and adverse events (AE) after each dose were generally mild to moderate and transient. Solicited AEs in Older Adults and Adults with Comorbidities were generally less frequent than in Healthy Adults. CoVLP+AS03 induced seroconversion in &gt;35% of subjects in each group after the first dose and in ~98% of subjects 21 days after the second dose. In all treatment groups, NAb levels were ~10-fold higher than those in a panel of convalescent sera. A significant minority (~20%) of subjects had evidence of a pre-existing IFN-γ response to the S protein and almost all subjects in all groups (&gt;88%) had detectable cellular responses (IFN-γ, IL-4 or both) at 21 days after the second dose. A Th1-biased response was most evident after the first dose and was still present after dose two. These data demonstrated that CoVLP+AS03 will likely be well-tolerated and highly immunogenic in adults ≥18 years of age with and without comorbidities.

<li><strong>Impact of Vaccine Prioritization Strategies on Mitigating COVID-19: An Agent-Based Simulation Study using an Urban Region in the United States</strong> -

Background Approval of novel vaccines for COVID-19 had brought hope and expectations, but not without additional challenges. One central challenge was understanding how to appropriately prioritize the use of limited supply of vaccines. This study examined the efficacy of the various vaccine prioritization strategies using the vaccination campaign underway in the U.S. Methods The study developed a granular agent-based simulation model for mimicking community spread of COVID-19 under various social interventions including full and partial closures, isolation and quarantine, use of face mask and contact tracing, and vaccination. The model was populated with parameters of disease natural history, as well as demographic and societal data for an urban community in the U.S. with 2.8 million residents. The model tracks daily numbers of infected, hospitalized, and deaths for all census age-groups. The model was calibrated using parameters for viral transmission and level of community circulation of individuals. Published data from the Florida COVID-19 dashboard was used to validate the model. Vaccination strategies were compared using a hypothesis test for pairwise comparisons. Results Three prioritization strategies were examined: a minor variant of CDC9s recommendation, an age-stratified strategy, and a random strategy. The impact of vaccination was also contrasted with a no vaccination scenario. The study showed that the campaign against COVID-19 in the U.S. using vaccines developed by Pfizer/BioNTech and Moderna 1) reduced the cumulative number of infections by 10% and 2) helped the pandemic to subside below a small threshold of 100 daily new reported cases sooner by approximately a month when compared to no vaccination. A comparison of the prioritization strategies showed no significant difference in their impacts on pandemic mitigation. Conclusions Even though vaccines for COVID-19 were developed and approved much quicker than ever before, their impact on pandemic mitigation was small as the explosive spread of the virus had already infected a significant portion of the population, thus reducing the susceptible pool. A notable observation from the study is that instead of adhering strictly to a sequential prioritizing strategy, focus should perhaps be on distributing the vaccines among all eligible as quickly as possible, after providing for the most vulnerable. As much of the population worldwide is yet to be vaccinated, results from this study should aid public health decision makers in effectively allocating their limited vaccine supplies.

<li><strong>Impact of the COVID-19 pandemic on mental health among Greek adults: a cross-sectional survey.</strong> -

The coronavirus disease 2019 (COVID-19) pandemic and mitigation measures based on social distancing are expected to have serious adverse effects on mental health. This cross-sectional study aimed to examine self-reported changes in the mental health status of Greek adults. The current study is a primary research conducted on Greek adults during the first wave of the epidemic (March to April 2020). A total of 527 individuals participated in an online survey using a validated questionnaire (State-Trait Anxiety Inventory-STAI and DASS-21). Results: The respondents had a moderate mental health status based on the following scores: STAI-S, 45.8; STAI-T, 40.7; depression, 4.6; anxiety, 3.1; and stress, 6.1. Women, younger respondents, those from lower income households, and those living in smaller apartments experienced increased depression, anxiety, and stress. Additionally, infection control practices during the COVID-19 pandemic such as the use of masks, gloves, and antiseptic can drastically decrease the prevalence of mental health illnesses. These findings can be used by the Greek State to reduce the effects of COVID-19 on the mental health of the population and protect socially vulnerable groups.

<li><strong>AUG-3387, a Human-Derived Monoclonal Antibody Neutralizes SARS-CoV-2 Variants and Reduces Viral Load from Therapeutic Treatment of Hamsters In Vivo</strong> -

Infections from the SARS-CoV-2 virus have killed over 4.6 million people since it began spreading through human populations in late 2019. In order to develop a therapeutic or prophylactic antibody to help mitigate the effects of the pandemic, a human monoclonal antibody (mAb) that binds to the SARS-CoV-2 spike protein was isolated from a convalescent patient following recovery from COVID-19 disease. This mAb, designated AUG-3387, demonstrates a high affinity for the spike protein of the original viral strains and all variants tested to date. In vitro pseudovirus neutralization and SARS-CoV-2 neutralization activity has been demonstrated in vitro. In addition, a dry powder formulation has been prepared using a Thin Film Freezing (TFF) process that exhibited a fine particle fraction (FPF) of 50.95 {+/-} 7.69% and a mass median aerodynamic diameter (MMAD) and geometric standard deviation (GSD) of 3.74 {+/-} 0.73 m and 2.73 {+/-} 0.20, respectively. The dry powder is suitable for delivery directly to the lungs of infected patients using a dry powder inhaler device. Importantly, AUG-3387, administered as a liquid by intraperitoneal injection or the dry powder formulation delivered intratracheally into Syrian hamsters 24 hours after intranasal SARS-CoV-2 infection, demonstrated a dose-dependent reduction in the lung viral load of the virus. These data suggest that AUG-3387 formulated as a dry powder demonstrates potential to treat COVID-19.

<li><strong>SARS-CoV-2 spike protein induces abnormal inflammatory blood clots neutralized by fibrin immunotherapy</strong> -

Blood clots are a central feature of coronavirus disease-2019 (COVID-19) and can culminate in pulmonary embolism, stroke, and sudden death. However, it is not known how abnormal blood clots form in COVID-19 or why they occur even in asymptomatic and convalescent patients. Here we report that the Spike protein from severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) binds to the blood coagulation factor fibrinogen and induces structurally abnormal blood clots with heightened proinflammatory activity. SARS-CoV-2 Spike virions enhanced fibrin-mediated microglia activation and induced fibrinogen-dependent lung pathology. COVID-19 patients had fibrin autoantibodies that persisted long after acute infection. Monoclonal antibody 5B8, targeting the cryptic inflammatory fibrin epitope, inhibited thromboinflammation. Our results reveal a procoagulant role for the SARS-CoV-2 Spike and propose fibrin-targeting interventions as a treatment for thromboinflammation in COVID-19.

Understanding the immune response to severe acute respiratory syndrome coronavirus (SARS-CoV-2) is critical to overcome the current coronavirus disease (COVID-19) pandemic. Efforts are being made to understand the potential cross- protective immunity of memory T cells, induced by prior encounters with seasonal coronaviruses, in providing protection against severe COVID-19. In this study we assessed T-cell responses directed against highly conserved regions of SARS- CoV-2. Epitope mapping revealed 16 CD8 + T-cell epitopes across the nucleocapsid (N), spike (S) and ORF3a proteins of SARS-CoV-2 and five CD8 + T-cell epitopes encoded within the highly conserved regions of the ORF1ab polyprotein of SARS-CoV-2. Comparative sequence analysis showed high conservation of SARS-CoV-2 ORF1ab T-cell epitopes in seasonal coronaviruses. Paradoxically, the immune responses directed against the conserved ORF1ab epitopes were infrequent and subdominant in both convalescent and unexposed participants. This subdominant immune response was consistent with a low abundance of ORF1ab encoded proteins in SARS-CoV-2 infected cells. Overall, these observations suggest that while cross- reactive CD8 + T cells likely exist in unexposed individuals, they are not common and therefore are unlikely to play a significant role in providing broad pre-existing immunity in the community.

<li><strong>A SARS-CoV-2 variant elicits an antibody response with a shifted immunodominance hierarchy</strong> -

Many SARS-CoV-2 variants have mutations at key sites targeted by antibodies. However, it is unknown if antibodies elicited by infection with these variants target the same or different regions of the viral spike as antibodies elicited by earlier viral isolates. Here we compare the specificities of polyclonal antibodies produced by humans infected with early 2020 isolates versus the B.1.351 variant of concern (also known as Beta or 20H/501Y.V2), which contains mutations in multiple key spike epitopes. The serum neutralizing activity of antibodies elicited by infection with both early 2020 viruses and B.1.351 is heavily focused on the spike receptor-binding domain (RBD). However, within the RBD, B.1.351-elicited antibodies are more focused on the “class 3” epitope spanning sites 443 to 452, and neutralization by these antibodies is notably less affected by mutations at residue 484. Our results show that SARS-CoV-2 variants can elicit polyclonal antibodies with different immunodominance hierarchies.

<li><strong>Restrictions of Rights and Freedoms during the COVID-19 Pandemic in the United Kingdom of Great Britain and Northern Ireland</strong> -

In the United Kingdom of Great Britain and Northern Ireland fundamental rights and freedoms are protected by the Human Rights Act 1998. Some of them were limited during a state of emergency declared on the basis of Civil Contingencies Act 2004 and, particularly relevant for the current COVID-19 crisis, Public Health (Control of Disease) Act 1984. The review of legal documents and literature indicates that the lockdown regulations mainly Health Protection (Coronavirus Restrictions) (England) Regulations 2020 adversely affected the everyday life of British citizens. This article discusses if those restrictions could have potentially interfered with some of the fundamental rights.

<li><strong>Effects of Age, Sex, Serostatus and Underlying Comorbidities on Humoral Response Post-SARS-CoV-2 Pfizer-BioNTech Vaccination: A Systematic Review</strong> -

With the advent of the Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) pandemic, several vaccines have been developed to mitigate its spread and prevent adverse consequences of the Coronavirus Disease 2019 (COVID-19). The mRNA technology is an unprecedented vaccine, usually given in two doses to prevent SARS-CoV-2 infections. Despite effectiveness and safety, inter-individual immune response heterogeneity has been observed in recipients of mRNA-based vaccines. As a novel disease, the specific immune response mechanism responsible for warding off COVID-19 remains unclear at this point. However, significant evidence suggests that humoral response plays a crucial role in affording immunoprotection and preventing debilitating sequelae from COVID-19. As such this paper focused on the possible effects of age, sex, serostatus, and comorbidities on humoral response (i.e., total antibodies, IgG and/or IgA) of different populations post-mRNA-based Pfizer-BioNTech vaccination. A systematic search of literature was performed through PubMed, Cochrane CENTRAL, and Google Scholar. Studies were included if they reported humoral response to COVID-19 mRNA vaccines. A total of 32 studies was identified and reviewed, and the percent difference of means of reported antibody levels were calculated for comparison. Findings revealed that older individuals, the male sex, seronegativity, and those with more comorbidities mounted less humoral immune response. Given these findings, several recommendations were proposed regarding the current vaccination practices. These include giving additional doses of vaccination for immunocompromised and elderly populations. Another recommendation is conducting clinical trials in giving a combined scheme of mRNA vaccines, protein vaccines, and vector-based vaccines.

Background: Managing high levels of severe COVID-19 in the acute setting can impact upon the quality of care provided to both affected patients and those requiring other hospital services. Mass vaccination has offered a route to reduce societal restrictions while protecting hospitals from being overwhelmed. Yet, early in the mass vaccination effort, the possible effect on future bed pressures remained subject to considerable uncertainty. This paper provides an account of how, in one healthcare system, operational decision-making and bed planning was supported through modelling the effect of a range of vaccination scenarios on future COVID-19 admissions. Methods: An epidemiological model of the Susceptible-Exposed-Infectious-Recovered (SEIR) type was fitted to local data for the one-million resident healthcare system located in South West England. Model parameters and vaccination scenarios were calibrated through a system-wide multi-disciplinary working group, comprising public health intelligence specialists, healthcare planners, epidemiologists, and academics. <li>Results: Achieving 95% vaccine uptake in adults by 31 July 2021 would not avert a third wave in autumn 2021 but would produce a median peak bed requirement approximately 6% (IQR: 1% to 24%) of that experienced during the second wave (January 2021). A two-month delay in vaccine rollout would lead to significantly higher peak bed occupancy, at 66% (11% to 146%) of that of the second wave. If only 75% uptake was achieved (the amount typically associated with vaccination campaigns) then the second wave peak for acute and intensive care beds would be exceeded by 4% and 19% respectively, an amount which would seriously pressure hospital capacity. Conclusion: Modelling provided support to senior managers in setting the number of acute and intensive care beds to make available for COVID-19 patients, as well as highlighting the importance of public health in promoting high vaccine uptake among the population. Forecast accuracy has since been supported by actual data collected following the analysis, with observed peak bed occupancy falling comfortably within the inter-quartile range of modelled projections.

Background: It is hypothesized that different levels of hormones especially serum testosterone level could explain the sex differences between men and women on the susceptibility and case fatality rate of COVID-19. However, traditional observational studies that support this hypothesis could not effectively establish the causal effects. Objective: Utilizing recently published genome-wide associations studies (GWAS) on serum Testosterone level and on COVID-19 related phenotypes, we sought to assess the causality through Mendelian Randomization (MR) analyses. We further applied a suite of statistical genomics methods to further explore the biological mechanisms. Results: We found that testosterone level is significantly associated with Covid-19 critical illness. All six MR methods yielded significant associations. There is no significant association between Testosterone and COVID-19 respiratory failure or COVID-19 susceptibility. Conclusion: Based on the GWAS currently available, we provide support for a causal role of Testosterone on COVID-19 critical illness. Nevertheless, we recognize that the COVID-19 susceptibility GWAS effort is still ongoing and there is no such strong locus as CCR5 for HIV discovered for COVID-19.

<li><strong>Heterologous SARS-CoV-2 Booster Vaccinations: Preliminary Report</strong> -

Background: While Coronavirus disease 2019 (Covid-19) vaccines are highly effective, breakthrough infections are occurring. Booster vaccinations have recently received emergency use authorization (EUA) for certain populations but are restricted to homologous mRNA vaccines. We evaluated homologous and heterologous booster vaccination in persons who had received an EUA Covid-19 vaccine regimen. Methods: In this phase 1/2 open-label clinical trial conducted at ten U.S. sites, adults who received one of three EUA Covid-19 vaccines at least 12 weeks prior to enrollment and had no reported history of SARS-CoV-2 infection received a booster injection with one of three vaccines (Moderna mRNA-1273 100-mcg, Janssen Ad26.COV2.S 5x10<sup>10</sup> virus particles, or Pfizer-BioNTech BNT162b2 30-mcg; nine combinations). The primary outcomes were safety, reactogenicity, and humoral immunogenicity on study days 15 and 29. Results: 458 individuals were enrolled: 154 received mRNA-1273, 150 received Ad26.CoV2.S, and 154 received BNT162b2 booster vaccines. Reactogenicity was similar to that reported for the primary series. Injection site pain, malaise, headache, and myalgia occurred in more than half the participants. Booster vaccines increased the neutralizing activity against a D614G pseudovirus (4.2-76-fold) and binding antibody titers (4.6-56-fold) for all combinations; homologous boost increased neutralizing antibody titers 4.2-20-fold whereas heterologous boost increased titers 6.2-76-fold. Day 15 neutralizing and binding antibody titers varied by 28.7-fold and 20.9-fold, respectively, across the nine prime-boost combinations. Conclusion: Homologous and heterologous booster vaccinations were well-tolerated and immunogenic in adults who completed a primary Covid-19 vaccine regimen at least 12 weeks earlier.

<li><strong>An objective systematic comparison of the most common adverse events of COVID-19 vaccines</strong> -

Background: Vaccination is an important tool in the fight against pandemics. However, the associated adverse events (AEs) may negatively impact the public perception of vaccines, therefore leading to decreased vaccination willingness. Interestingly, pharmacovigilance data of the three COVID-19 vaccines with a two-dose schedule approved in the EU (AstraZeneca, BioNTech and Moderna) already revealed country-specific differences in their safety profile early on (as of February 2021), at a time when the accumulated occurrence of specific AEs was not yet known. In the safety outcome assessment presented here, we aimed to establish whether these country-specific differences in pharmacovigilance data could be explained by differences in the frequency of AEs as reported in the respective approval studies of each vaccine. Methods: A systematic search was performed to identify all publications regarding the randomized controlled trials (RCTs) of two-dose vaccines approved in the EU (AstraZeneca, BioNTech and Moderna), including regulatory reports and journal articles. All obtained safety data was manually entered into an SQL database. In order to enable the comparability among the data, the solicited AEs for all vaccines (i.e. those AEs actively sought after vaccination) were investigated. The data was standardized to promote comparability and overcome data heterogeneity and complexity. Findings: Twelve documents regarding the RCTs for the three COVID-19 vaccines with a two-dose schedule approved in the EU (AstraZeneca, BioNTech and Moderna) were included in the safety outcome analysis. The entire safety data compiled in the SQL database amounted to 66 different study arms. The data structure revealed 13 different age thresholds or ranges and three different data sets regarding doses (first dose vs. second dose vs. all doses). After standardization and identification of subgroups, the analyses demonstrated that the highest rates of AEs occur after the first dose with the AstraZeneca vaccine, whereas with Moderna and BioNTech most AEs occur after the second dose. Astonishingly, the absolute frequencies of each AE after the first AstraZeneca dose correspond to those of the second dose of the mRNA vaccines (BioNTech and Moderna). Reversely, the absolute frequencies of the same AEs after the second AstraZeneca dose correspond to those of the first dose with the mRNA vaccines. The most common AEs with any vaccine were fatigue, headache and myalgia. Moreover, middle-aged subjects (18 to 55 years) had more side effects than older individuals (&gt; 55 years), an observation that persisted among vaccines. Interpretation: This is the first indirect comparison of these vaccines that uses all available RCT data. The absolute frequency of each AE is similar between the first AstraZeneca dose and the second dose of BioNTech or Moderna; their occurrence was thus independent of platform (vector or mRNA) or the vaccine itself. This assessment demonstrates that the varying frequencies of AEs reported in early pharmacovigilance data for the vaccines in distinct countries, at a time when the accumulated occurrence of specific AEs with certain vaccines was not yet known, cannot be explained by different frequencies being reported in the respective RCTs. Conclusion: The approach presented here could help to objectify future discussions on vaccine preferences. Therefore, it may serve as basis for future public awareness campaigns and may also allow the comparison of vaccine performance in different subgroups (e.g. virus variants, high-risk patients). This approach may also be applied to a broad range of other challenges across the R&amp;D process and various disease categories.

You may not have seen <em>Succession</em>, but maybe youve heard about <a href="https://www.youtube.com/watch?v=6dUDQTc-9kM">Kendall Roys supremely cringey rap</a>, or the line “<a href="https://www.youtube.com/watch?v=ezAlySFluEk">You cant make a Tomelette without breaking some Greggs</a>.”
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="HvvdNh">
Maybe youve seen tweets about how much <a href="https://twitter.com/abbotonline/status/1178428548744392707">the theme song</a> is a banger. Or heard brilliant comedian Demi Adejuyigbes “<a href="https://www.youtube.com/watch?v=e-6K2CjJ6dk">Kiss from Daddy</a>,” which adds ridiculous lyrics to said theme song. Perhaps the words “boar on the floor” mean something to you?
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="VVee9L">
If youre not a fan of HBOs darkly comedic primetime soap, you may not be aware of all of the above, but at least one of any number of <em>Succession</em> memes, jokes, and flash points has probably pierced your social media bubble, even if you were completely baffled by it. The show has slowly but surely wormed its way into the hearts of People Who Talk About TV Online, and that has allowed it to become the prestige drama of the moment, winning near-universal critical praise and nine Emmys across its first two seasons.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="4p8z9I">
A lot of that success is due to how easy it is to boil down the show to its most immediately hilarious jokes, awkward moments, and jaw-dropping plot twists. But I would argue that just as much of it stems from how ably <em>Succession</em> incorporates themes our culture is obsessed with right now. In its portrayal of a broken family that is making the world a little bit worse every single day, the series speaks to an era where billionaires <a href="https://www.washingtonpost.com/technology/2018/11/26/elon-musk-says-he-
will-probably-move-mars/">openly muse about moving to a new planet</a> if we irreparably screw this one up.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="8fAMs4">
<em>Succession</em> is many, many things, but the series has five specific modes that have made it such a darling of people who care about TV. Those modes illustrate just how completely the series has encapsulated our modern world.
</p>
<h3 id="rSqeup">
Mode 1: A lacerating portrayal of wealths corruptive power
</h3>

If you asked even the most casual viewer of <em>Succession</em> to identify its core theme, “wealth corrupts those who hold it” (or something to that effect) would almost surely be part of their answer. Wealth is everywhere in <em>Succession</em>, but its rarely displayed ostentatiously. The characters are so used to having obscene amounts of money, they seem unaware that not everyone enjoys the same levels of comfort.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="B5qf6i">
The Roy family at the center of <em>Succession</em> is a kind of portmanteau of the Murdoch family (owners of the News Corp empire, encompassing Fox News, the Fox Broadcasting Company, and numerous publications) and the Redstone family (majority shareholder of the ViacomCBS empire, which includes numerous TV networks as well as Paramount Pictures). Paterfamilias Logan Roy (Brian Cox) is past his prime, and in the series very first episode, he almost dies. But hes very good at yelling, and everybody around him is so terrified of his whims that he tends to get his way, even when hes recovering from a debilitating stroke.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="rjbeya">
<em>Succession</em>s first episode almost immediately established just how divorced the Roys are from reality. In one of its most memorable sequences, Roman (Kieran Culkin), the sleazy and opportunistic youngest brother of the family, offers a working-class kid $1 million if he can hit a home run. He cant, and Roman chuckles at the whole ordeal while a few of the lampreys who have attached themselves to the Roy familys belly go to pay off the kids parents so that they dont tell anybody anything.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="YVyK6V">
Broadly speaking, <em>Succession</em> lives within the “primetime soap about a rich family” subgenre, but rather than reveling in the opulence and decadence of extreme wealth in the fashion of <em>Dynasty</em> or <em>Dallas</em>, <em>Succession</em> makes everything feel a little gross. The show is careful to never present wealth as a prison that holds the characters back — they could always just buy six jets and move to Antarctica — but it never loses sight of the fact that <em>we</em>, the viewers, live in a socioeconomic prison built in large part by people who have more money than they could ever reasonably spend.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="oCPdIU">
Every so often, a character on <em>Succession</em> suggests the Roys are destroying the planet, and the show never offers a response other than, “Yes, almost certainly.” Theres no reason for anybody to have so much money, and the Roys wealth serves primarily to make them squabble endlessly with each other over it. <em>Succession</em> makes you empathize with billionaires without ever making you like them. Its a neat trick.

Mode 2: An extremely dark comedy
</h3>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="XSnDCW">
Make no mistake: <em>Succession</em> is a uniquely despairing drama about a family (and a planet) circling the drain. But the reason its watchable at all is that its also one of TVs funniest shows.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="MnnasH">
It comes by its comedic bona fides honestly. Creator and showrunner Jesse Armstrong was known predominantly for comedy before taking this turn toward the dramatic, having co-created the legendary British series <em>Peep Show</em> and written for the British political satire <em>The Thick of It</em>. He even earned an Oscar nomination for co-writing the screenplay of <em>In the Loop</em>, the film adaptation of <em>The Thick of It</em>. His keen sense of satire is to <em>Succession</em>s great benefit.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="ua76LY">
Consider, “You cant make a Tomelette without breaking some Greggs,” a joke from the <a href="https://www.vox.com/culture/2019/10/6/20897073/succession-season-2-episode-9-dc-recap-winners-losers-tom-
greg">penultimate episode of season two</a>. Its part of an email read by a congressman at a Senate subcommittee hearing investigating the Roy familys shady handling of sexual assault allegations tied to their cruise line.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="aP7aLC">
The joke, then, is part of something uniquely dark and horrible, but its also just such a good joke. It skillfully plays off the fact that Roy family son-in-law Tom Wambsgans (Matthew Macfadyen) and gangly nephew/cousin Greg (Nicholas Braun) are the shows Rosencrantz and Guildenstern, lurking around the storys edges to have comedic side plots that occasionally intersect with the action.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="Xz6SvX">
“Tomelette” and “Greggs” isnt just a funny combination of puns. It underscores the characters roles within both the show and this storyline, as well as the callousness with which the Roy family treats almost every serious wrong committed by Waystar Royco, the media conglomerate the family owns. It might be perfect.

Beyond oddly significant puns, <em>Succession</em> boasts a thorough command of a wide range of comedic styles. Its as adroit with cringe comedy as it is with wordplay, and it allows its characters to be clever without turning everyone into a quip machine. I laugh many, many times watching each episode of <em>Succession</em>, without ever feeling like the show undercuts its storytelling with absurdity. Its a neat trick.

Mode 3: An accurate, depressing media satire

Early in season two, someone in my office got their hands on a <a href="https://twitter.com/emilyvdw/status/1170035215453999104">baseball cap emblazoned with Vaulter</a>, the fake version of Gawker that the Roy family destroys across the course of <em>Succession</em>s first two seasons. When I posted a photo of myself wearing it on social media, seemingly every journalist I know reached out to ask where Id gotten it. They cant have it. Its mine.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="lTLlUd">
I share this anecdote to elucidate the degree to which <em>Succession</em> resonates with media types: Its portrayal of how the Roys blithely destroy everything they touch to protect their own interests emphasizes just how perilous journalism has become at this point in history. Profit margins are razor thin, and increased media consolidation means that independent voices within the industry are largely marginalized or snuffed out. </p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="wPXC8J">
Of all the modes of <em>Succession</em>, “media satire” is the one that probably speaks to the smallest number of people. But at a certain point, if everybody who works in media is watching a show and treating it like its the only show on TV that matters, a self-fulfilling prophecy emerges. <em>Succession</em> is popular beyond media bubbles, but its popularity within media bubbles certainly didnt hurt it with regard to getting early attention.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="OVxwQE">
Im not sure that <em>Succession</em> has as much to say about the media as it does many other topics, but maybe the simple act of saying, “This seems like a shitty situation,” is enough in an era when the economic pressures that threaten to suffocate the entire media industry add up to a shitty situation.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="5zCvhc">
I do find its portrayal of the ways media titans use their companies mostly to create realities where they never need to feel challenged or upbraided appropriately chilling. Logan Roy is really good at yelling at people until reality bends to become what he wants it to be. His media empire is an extension of that.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="GXaRgc">
On the whole, media is even more incidental to <em>Succession</em> than, say, advertising was to <em>Mad Men</em> or chemistry was to <em>Breaking Bad</em> by merely offering a vague milieu in which the series can take place. Still, whenever <em>Succession</em> drops you into the offices of Waystar Royco, it almost always nails what its like to work there. Its a neat trick.
</p>
<h3 id="HG8Q7p">
Mode 4: An acutely observant exploration of internecine family squabbling
</h3>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="NcluuT">
If <em>Succession</em>s most obvious ancestral genre is the primetime soap about rich people, it also has a healthy dose of the family drama baked into its DNA. (Then again, most primetime soaps, from <em>Dallas</em> to <em>Empire</em>, are sneakily family dramas.) The core of the series rests atop the ever-shifting relationships among the four Roy siblings, especially as they compete to win their fathers approval and affection.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="WqrE6k">
The series is at its sharpest when it focuses on the three youngest Roy siblings — depressive would-be heir apparent Kendall (Emmy winner Jeremy Strong), slinky cad Roman, and desperate girlboss Shiv (Sarah Snook). The three have an older half- brother, Connor (Alan Ruck), who has a different mother from them, but Connor is more of a comic-relief character, at least so far.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="shd5CG">
<em>Succession</em> is just <em>so smart</em> about the ways siblings both support and undercut each other, sometimes in the same moment. Tentative alliances form among all four of the Roy children, sometimes in opposition to Logan and sometimes in support of him, and the series digs into how these four particular people are the only ones who can know what its like to be raised by Logan Roy.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="2qf9PS">
But the series portrayal of complicated family dynamics extends beyond the siblings. Logans current wife Marcia (Hiam Abbass) emerges as a key player in her own right across <em>Succession</em>s first two seasons, while Tom and Greg are more peripheral family members who perpetually seem to be stuck at the kids table.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="JkpkxU">
<em>Succession</em> also broadens its portrayal of family in a metaphorical direction, by underlining the poisonous nature of corporations that insist all their employees are part of a giant family. Waystar Royco might be a family, but its just as fucked up as the Roy family. (Keep an eye on J. Smith-Cameron as Gerri, who starts out as a minor player in season one but becomes more and more important as the show continues.)
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="SBIs0B">
Beyond even Waystar Roycos own staff, the series pulls in a handful of other rich and powerful families for comparison and contrast against the Roy family. Notably, season two introduces the Pierces, a loose riff on the Sulzbergers (owners of the New York Times) and the sort of old-money liberals Logan Roy was so clearly reacting against when he was young and hungry. <p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="nrolqi">
If I had to choose just one of its modes to explain why <em>Succession</em> has taken off the way it has, Id choose this one. The shows finely tuned understanding of complicated family dynamics means that you can surely find one or two details in its portrayal of the Roy family that resonate with your own life — and might leave you thankful your family isnt as messed up as the Roys. Its a neat trick.
</p>

But on the “acutely observed family dynamics” front, the series ultimately goes one level deeper than even that.

Mode 5: A psychologically trenchant excavation of the legacy of abuse

<em>Succession</em> often portrays Logan Roy as a Shakespearean figure, a little like Lear bellowing at the skies to obey him and not being heard. But because it places his titanic rage within settings that viewers are at least somewhat familiar with — boardrooms and dining rooms — it becomes easier, as the show goes on, to realize Logan is a terrible, abusive parent. At one point in season two, he physically strikes Roman for a minor slight, and all the other siblings either flinch instinctively or stare, dead-eyed, as it happens.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="GHFr9V">
Logan Roy has always gotten his way. Hes built his life around the idea that because he thinks something is true, it must indeed be true, and he has built a media empire around similar principles of telling people what they want to hear and not what they actually need to know. He shouts and he blusters and he snaps at people, and if he doesnt get what he wants, anybody close to him feels the effects of his ire.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="9hk0uG">
In Logans case, those who are closest to him have always been his children. <em>Succession</em>s first season hinted at the depths of sadness to which Logan has driven his kids, and then the second season made that sadness both the subtext and text of nearly every scene. The Roy siblings want so badly to be loved by a man who is only capable of trying to bludgeon them (occasionally literally) into a form that is more pleasing to him, which is to say a form that is more subservient to him.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="srLYo9">
Yes, <em>Succession</em> is funny; yes, its about wealth; and, yes, its really smart about the media. But for my money, the show is smartest about what it means to have an abusive parent. The longer <em>Succession</em> runs, the more Logans monstrousness is revealed as critical to the formation of his children. They long desperately to be loved, but what they get is mostly a sense that they have no control over anything in their lives. They spread that lack of control outward to everyone around them. They are a ruling class that does not understand what it means to rule, much less how much their ignorance damages everyone they come into contact with, because all they have known is damage.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="uAbtJH">
To be alive in the 2020s is to become more and more aware of how deep the rot goes, to realize how unlikely any of us is to turn the car around before it goes over the cliff. We exist in a world full of men who believe that if they shout something loud enough, it is true — and who take extreme<strong> </strong>offense to the idea that maybe they are wrong, no matter how gently we break it to them. <em>Succession</em> captures that feeling, and … I dont know if its a neat trick, but it feels like right now more than any other show on television.

Chaillan, a 37-year-old tech entrepreneur, added that cyber defenses at many government agencies are at “kindergarten level,” and that companies like Google were doing the US a disservice by not working with the military more on AI, since Chinese companies were making a “massive investment” in AI without getting all hung up on the ethics of it all. And while quitting your job because America has already lost the AI race is a bit dramatic, Chaillan isnt the only one whos concerned about Chinas dominance in this arena.

A growing number of leaders in Washington and Silicon Valley are worried about the US falling behind in the race to AI supremacy. Congressional hearings on the future of AI have been going on <a href="https://link.recode.net/click/25331141.52099/aHR0cHM6Ly93d3cuY29tbWVyY2Uuc2VuYXRlLmdvdi8yMDE2LzExL2NvbW1lcmNlLWFubm91bmNlcy1maXJzdC1hcnRpZmljaWFsLWludGVsbGlnZW5jZS1oZWFyaW5n/608c6cd77e3ba002de9a4c0dB7fd58196">since 2016</a>, and Chaillan said he plans to testify in some upcoming ones. Earlier this year, the National Security Commission on AI, a project chaired by former Google CEO Eric Schmidt, also boldly declared that China <a href="https://link.recode.net/click/25331141.52099/aHR0cHM6Ly93d3cuY25iYy5jb20vMjAyMS8wMy8wMi91cy1ub3QtcHJlcGFyZWQtdG8tZGVmZW5kLW9yLWNvbXBldGUtaW4tYWktZXJhLXNheXMtZXJpYy1zY2htaWR0LWdyb3VwLmh0bWw/608c6cd77e3ba002de9a4c0dB3073e050">is poised to surpass the US</a> as the worlds “AI superpower.” In a statement signed by Elon Musk, Jack Dorsey, and Stephen Hawking, among thousands of scientists, the commission said, “AI technology has reached a point where the deployment of such systems is — practically if not legally — feasible within years, not decades, and the stakes are high: autonomous weapons have been described as the third revolution in warfare, after gunpowder and nuclear arms.”

We can all agree that nobody wants China to invent a real-world version of Skynet, the all-powerful AI that <a href="https://link.recode.net/click/25331141.52099/aHR0cHM6Ly93d3cueW91dHViZS5jb20vd2F0Y2g_dj1fV2xzZDltbGppVQ/608c6cd77e3ba002de9a4c0dB971de2c5">takes over the planet</a> in the <em>Terminator </em>movies. But we dont want the US to do that either. And what does the finish line in this AI race actually look like? Does the US really want to win at all costs?
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="GoXoHH">
For years, pundits have been <a href="https://link.recode.net/click/25331141.52099/aHR0cHM6Ly9mb3JlaWducG9saWN5LmNvbS8yMDE3LzExLzAzL3RoZS1uZXh0LXNwYWNlLXJhY2UtaXMtYXJ0aWZpY2lhbC1pbnRlbGxpZ2VuY2UtYW5kLWFtZXJpY2EtaXMtbG9zaW5nLXRvLWNoaW5hLw/608c6cd77e3ba002de9a4c0dB2cb6a031">comparing the AI race to the space race </a>— and warning that the US is losing it. Its a handy analogy, since it helps Americans put current conflicts with countries like China and Russia into the familiar context of the Cold War. Many have argued that weve found ourselves <a href="https://link.recode.net/click/25331141.52099/aHR0cHM6Ly9naXptb2RvLmNvbS9tZWV0LXRoZS1uZXctY29sZC13YXItc2FtZS1hcy10aGUtb2xkLWNvbGQtd2FyLTE2NjAzMjgyNzA/608c6cd77e3ba002de9a4c0dB3bb71c88">in a second Cold War</a> and that the country that wins the AI race will take the throne as the dominant superpower. But the AI revolution isnt just about fighting wars or geopolitical dominance. What were racing to build will transform almost every aspect of our lives, from how we run businesses to how we process information to how we get around.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="zlVAMS">
So its imperative that the US is thoughtful about quickly charging into a future filled with autonomous cars, boundless data collection, and full-time surveillance. These are the applications that next-generation AI will enable, and if a small group of powerful tech companies and/or the US military pushes for innovation without <a href="https://link.recode.net/click/25331141.52099/aHR0cHM6Ly9oYnIub3JnLzIwMjEvMDkvYWktcmVndWxhdGlvbi1pcy1jb21pbmc/608c6cd77e3ba002de9a4c0dBdfcbc236">putting the proper guard rails in place</a>, this world-changing technology could lead to some grim unintended consequences. President Biden <a href="https://link.recode.net/click/25331141.52099/aHR0cHM6Ly93d3cucG9saXRpY28uY29tL25ld3MvMjAyMS8wMy8wMi9jaGluYS11cy1ldXJvcGUtYWktcmVndWxhdGlvbi00NzIxMjA/608c6cd77e3ba002de9a4c0dB02b5580c">called for the US and Europe to work together</a> on developing new technology responsibly in a February speech at the Munich Security Conference.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="In1VQQ">
“We must shape the rules that will govern the advance of technology and the norms of behavior in cyberspace, artificial intelligence, biotechnology so that they are used to lift people up, not used to pin them down,” Biden said. “We must stand up for the democratic values that make it possible for us to accomplish any of this, pushing back against those who would monopolize and normalize repression.
</p>
<p data-aos="fade-left" data-aos-anchor-placement="bottom-bottom" id="aa53cE">
You could also look to present-day China to see what the near future of a more AI-centric society might look like. As Kai-Fu Lee argues in his book <a href="https://link.recode.net/click/25331141.52099/aHR0cHM6Ly93d3cuYW1hem9uLmNvbS9BSS1TdXBlcnBvd2Vycy1DaGluYS1TaWxpY29uLVZhbGxleS9kcC8xMzI4NTQ2MzlYLz9vdHM9MSZhc2NzdWJ0YWc9W11kdltwXWNqbjNsNHA4dDAwNHZyMXlldGNtM2pkZzlbaV1Udm5UeWtbdV0xW3Rdd1tyXWdvb2dsZS5jb21bZF1EW3pdbSZ0YWc9ZGV2ZWxvcGluZ255bWFn/608c6cd77e3ba002de9a4c0dBf2eaa65d"><em>AI Superpowers: China, Silicon Valley, and the New World Order</em></a>, China has been more aggressive at implementing AI breakthroughs, especially in <a href="https://link.recode.net/click/25331141.52099/aHR0cHM6Ly93d3cuZnQuY29tL2NvbnRlbnQvMmYyOTVhOWUtNWY5Ni0xMWU5LWIyODUtM2FjZDVkNDM1OTll/608c6cd77e3ba002de9a4c0dB7ee3414a">surveillance and data collection applications</a>, thanks in part to government support and a lack of oversight thats let some tech companies there leapfrog the competition and dominate entire industries. WeChat and its parent company, Tencent, are perfect examples of this. On WeChat, privacy <a href="https://link.recode.net/click/25331141.52099/aHR0cHM6Ly9mb3VuZGF0aW9uLm1vemlsbGEub3JnL2VuL3ByaXZhY3lub3RpbmNsdWRlZC93ZWNoYXQv/608c6cd77e3ba002de9a4c0dB582dfcf6">does not seem to be a priority</a>, but the vast quantities of data the app can collect are certainly helpful for training AI.
</p>

For the rest of us, the rise of AI shouldnt feel like a race against China. Its more like a high-stakes poker game.
